    The World Wide Web is an amazing place that people's lives more and more rely on. Especially, for the young generation, they spend a significant amount of their play and study time using the Internet. Many tools have been developed to help the educational users in finding educational resources. These tools include various search engines. Web directories and educational domain gateways. Nevertheless, these systems have many weaknesses that made them unsuitable for the specific search needs of the learners. The research presented in this thesis describes the development of the Web-course search engine, which is a friendly, efficient and accurate helper for the learners to get what they want in the vast Internet ocean. The most attractive feature of this system is that the system uses one universal language, which lets the searchers and the resources "communicate" with each other. Then the learner searchers can find the Web-based educational resources that are most fit to their needs and course providers can provide all necessary information about their courseware. This universal language is one widely acceptable Metadata standard. Following the Metadata standard, the system collects exact information about educational resources, provides adequate search parameters for search and returns evaluative results. By using the Web-course search engine, the learners and the other educational users are able to find useful, valuable and related educational resources more effectively and efficiently. Some improvement suggestions of the search mechanism in the World Wide Web have been brought forward for the future research as a result of this project

    LAUN Improved StarGAN for Facial Emotion Recognition

    In the field of facial expression recognition, deep learning is extensively used. However, insufficient and unbalanced facial training data in available public databases is a major challenge for improving the expression recognition rate. Generative Adversarial Networks (GANs) can produce more one-to-one faces with different expressions, which can be used to enhance databases. StarGAN can perform one-to-many translations for multiple expressions. Compared with original GANs, StarGAN can increase the efficiency of sample generation. Nevertheless, there are some defects in essential areas of the generated face, such as the mouth and the fuzzy side face image generation. To address these limitations, we improved StarGAN to alleviate the defects of images generation by modifying the reconstruction loss and adding the Contextual loss. Meanwhile, we added the Attention U-Net to StarGAN's generator, replacing StarGAN's original generator. Therefore, we proposed the Contextual loss and Attention U-Net (LAUN) improved StarGAN. The U-shape structure and skip connection in Attention U-Net can effectively integrate the details and semantic features of images. The network's attention structure can pay attention to the essential areas of the human face. The experimental results demonstrate that the improved model can alleviate some flaws in the face generated by the original StarGAN. Therefore, it can generate person images with better quality with different poses and expressions. The experiments were conducted on the Karolinska Directed Emotional Faces database, and the accuracy of facial expression recognition is 95.97%, 2.19% higher than that by using StarGAN. Meanwhile, the experiments were carried out on the MMI Facial Expression Database, and the accuracy of expression is 98.30%, 1.21% higher than that by using StarGAN. Moreover, experiment results have better performance based on the LAUN improved StarGAN enhanced databases than those without enhancement

    Sox10+ adult stem cells contribute to biomaterial encapsulation and microvascularization.

    Implanted biomaterials and biomedical devices generally induce foreign body reaction and end up with encapsulation by a dense avascular fibrous layer enriched in extracellular matrix. Fibroblasts/myofibroblasts are thought to be the major cell type involved in encapsulation, but it is unclear whether and how stem cells contribute to this process. Here we show, for the first time, that Sox10+ adult stem cells contribute to both encapsulation and microvessel formation. Sox10+ adult stem cells were found sparsely in the stroma of subcutaneous loose connective tissues. Upon subcutaneous biomaterial implantation, Sox10+ stem cells were activated and recruited to the biomaterial scaffold, and differentiated into fibroblasts and then myofibroblasts. This differentiation process from Sox10+ stem cells to myofibroblasts could be recapitulated in vitro. On the other hand, Sox10+ stem cells could differentiate into perivascular cells to stabilize newly formed microvessels. Sox10+ stem cells and endothelial cells in three-dimensional co-culture self-assembled into microvessels, and platelet-derived growth factor had chemotactic effect on Sox10+ stem cells. Transplanted Sox10+ stem cells differentiated into smooth muscle cells to stabilize functional microvessels. These findings demonstrate the critical role of adult stem cells in tissue remodeling and unravel the complexity of stem cell fate determination

    Effects of Altitude on Fire Smoke Diffusion in Semi-Lateral Smoke Exhaust Highway Tunnels

    This paper aims to study the effects of altitude and the size of smoke outlet on fire smoke diffusion and discharge in semi-lateral smoke exhaust highway tunnels. At first, simulations of semi-lateral smoke exhaust highway tunnels were carried out in FDS (Fire Dynamics Simulator), then the distribution laws of temperature, CO concentration, smoke mass flow, and visibility in the tunnel under the conditions of different altitudes and smoke outlet areas were analyzed to figure out the effects of altitude and size of smoke outlet on fire smoke diffusion and discharge in the said tunnels. The results suggest that, in case of the same fire source power, the velocity of smoke diffusion increases with the altitude; for curves of different altitudes, the tunnel roof temperature features are basically the same, that is, the higher the altitude, the higher the temperature at the tunnel roof. When the fire source power is 20 MW, the smoke mass flow at the smoke outlet decreases with the increase of altitude, but the CO concentration grows with it, indicating that the smoke exhaust efficiency is higher in high-altitude areas. When the altitude reaches 4200 m and the fire source power is 20 MW, with the increase of smoke outlet area, the smoke discharge effect of the tunnel shows an upward trend, taking both the smoke discharge effect and economy into consideration; the smoke outlet should take a size of 4 x 3 m

    Connections Between Connexins, Calcium, and Cataracts in the Lens

    There is a good deal of evidence that the lens generates an internal micro circulatory system, which brings metabolites, like glucose, and antioxidants, like ascorbate, into the lens along the extracellular spaces between cells. Calcium also ought to be carried into the lens by this system. If so, the only path for Ca2+ to get out of the lens is to move down its electrochemical gradient into fiber cells, and then move by electrodiffusion from cell to cell through gap junctions to surface cells, where Ca-ATPase activity and Na/Ca exchange can transport it back into the aqueous or vitreous humors. The purpose of the present study was to test this calcium circulation hypothesis by studying calcium homeostasis in connexin (Cx46) knockout and (Cx46 for Cx50) knockin mouse lenses, which have different degrees of gap junction coupling. To measure intracellular calcium, FURA2 was injected into fiber cells, and the gradient in calcium concentration from center to surface was mapped in each type of lens. In wild-type lenses the coupling conductance of the mature fibers was ∼0.5 S/cm2 of cell to cell contact, and the best fit to the calcium concentration data varied from 700 nM in the center to 300 nM at the surface. In the knockin lenses, the coupling conductance was ∼1.0 S/cm2 and calcium varied from ∼500 nM at the center to 300 nM at the surface. Thus, when the coupling conductance doubled, the concentration gradient halved, as predicted by the model. In knockout lenses, the coupling conductance was zero, hence the efflux path was knocked out and calcium accumulated to ∼2 μM in central fibers. Knockout lenses also had a dense central cataract that extended from the center to about half the radius. Others have previously shown that this cataract involves activation of a calcium-dependent protease, Lp82. We can now expand on this finding to provide a hypothesis on each step that leads to cataract formation: knockout of Cx46 causes loss of coupling of mature fiber cells; the efflux path for calcium is therefore blocked; calcium accumulates in the central cells; at concentrations above ∼1 μM (from the center to about half way out of a 3-wk-old lens) Lp82 is activated; Lp82 cleaves cytoplasmic proteins (crystallins) in central cells; and the cleaved proteins aggregate and scatter light

    Dominant cataracts result from incongruous mixing of wild-type lens connexins

    Gap junctions are composed of proteins called connexins (Cx) and facilitate both ionic and biochemical modes of intercellular communication. In the lens, Cx46 and Cx50 provide the gap junctional coupling needed for homeostasis and growth. In mice, deletion of Cx46 produced severe cataracts, whereas knockout of Cx50 resulted in significantly reduced lens growth and milder cataracts. Genetic replacement of Cx50 with Cx46 by knockin rescued clarity but not growth. By mating knockin and knockout mice, we show that heterozygous replacement of Cx50 with Cx46 rescued growth but produced dominant cataracts that resulted from disruption of lens fiber morphology and crystallin precipitation. Impedance measurements revealed normal levels of ionic gap junctional coupling, whereas the passage of fluorescent dyes that mimic biochemical coupling was altered in heterozygous knockin lenses. In addition, double heterozygous knockout lenses retained normal growth and clarity, whereas knockover lenses, where native Cx46 was deleted and homozygously knocked into the Cx50 locus, displayed significantly deficient growth but maintained clarity. Together, these findings suggest that unique biochemical modes of gap junctional communication influence lens clarity and lens growth, and this biochemical coupling is modulated by the connexin composition of the gap junction channels

    Whole-genome sequencing analysis of Klebsiella aerogenes among men who have sex with men in Guangzhou, China

    Klebsiella aerogenes is a common infectious bacterium that poses a threat to human health. Nevertheless, there are limited data on the population structure, genetic diversity, and pathogenicity of K. aerogenes, especially among men who have sex with men (MSM). The present study aimed to clarify the sequence types (STs), clonal complexes (CCs), resistance genes, and virulence factors of popular strains. Multilocus sequence typing was used to describe the population structure of K. aerogenes. The Virulence Factor Database and Comprehensive Antibiotic Resistance Database were used to assess the virulence and resistance profiles. In this study, next-generation sequencing was performed on nasal swabs specimens collected in an HIV Voluntary Counseling Testing outpatient department in Guangzhou, China, from April to August 2019. The identification results showed that a total of 258 K. aerogenes isolates were collected from 911 participants. We found that the isolates were most resistant to furantoin (89.53%, 231/258) and ampicillin (89.15%, 230/258), followed by imipenem (24.81%, 64/258) and cefotaxime (18.22%, 47/258). The most common STs in carbapenem-resistant K. aerogenes were ST4, ST93, and ST14. The population has at least 14 CCs, including several novel ones identified in this study (CC11-CC16). The main mechanism of drug resistance genes was antibiotic efflux. Based on the presence of the iron carrier production genes irp and ybt, we identified two clusters according to virulence profiles. In cluster A, CC3 and CC4 carry the clb operator encoding the toxin. Increased monitoring is needed for the three main ST type strains carried by MSM. The main clone group CC4 has a large number of toxin genes, and it spreads among MSM. Caution is needed to prevent further spread of this clone group in this population. In sum, our results may provide a foundation for the development of new therapeutic and surveillance strategies for treating MSM
